- A mixture of sample (e.g., plasma), internal standard, and buffer solution to adjust pH is prepared.
- This aqueous mixture is then added to the dry particle bed (no conditioning or pretreatment of the bed is necessary).
The mixture is allowed to partition for about 3-5 minutes on the particle surface under gravity flow.
- The analyte in aqueous solution is now spread out among the particles and occupies a high surface area. A hydrophobic filter at the bottom of each well prevents the aqueous phase from breaking through into the collection vessel placed underneath. Organic solvent is then added to the wells.
- The analyte will partition from the adsorbed aqueous phase into the organic solvent as this solvent slowly flows through the particle bed under gravity.
- The organic eluate is collected in wells or tubes underneath the tips of the plate. A second addition of organic solvent is performed and the combined eluates are evaporated to dryness. The residue is reconstituted in a mobile phase compatible solution and an aliquot is injected into the chromatographic system for analysis.
